併用黃耆萃取物對atenolol降血壓作用的影響

本計劃的主要目的為探討黃耆(Astragalus membranaceus)萃取物對β-blocker類藥物(atenolol)對先天性高血壓鼠的降血壓效果的作用,包括短期以及長期的影響,是否有加成或拮抗的作用。實驗方法為以先天性高血壓鼠為實驗模型來模擬高血壓的形成,觀察黃耆萃取物及g-aminobutyric acid (黃耆精抽物)對atenolol降血壓作用短期及長期影響。以正常血壓鼠WKY作對照組。此外為了了解其中交互作用的機轉,本實驗在併用黃耆萃取物或g-aminobutyric和atenolol後,觀測其血管反應性、血漿中nitrate及MCP-1濃度、胸主動脈superoxide產生量以及胸主動脈中eNOS及iNOS蛋白質及核酸(mRNA)表現量變化,以其能更了解黃耆萃取物及g-aminobutyric acid (黃耆精抽物)的藥理作用以及與atenolol之交互作用機轉,作為臨床醫師用藥的參考。

The influence of Astragalus Membranaceous on the antihypertencive effect of atenolol in spontaneously hypertensive rats

The aim of this study was to investigate the influence of Astragalus membranaceus on the anti-hypertensive effect of β-blocker drug, such as atenolol, in spontaneously hypertensive rats. In this study, the short- and long-term influences were determined. The spontaneously hypertensive rats were chosen to simulate the development of hypertension. The influences of the extraction of Astragalus membranaceus, and g-aminobutyric acid (a chemical compound of Astragalus membranaceus), on the short- and long-term effects of atenolol in SHR and WKY were observed. In addition, to further clarify the mechanisms involved, the vascular reactivity determination, the plasma nitrate and MCP-1levels, the superoxide production of aorta, and the eNOS (endothelial nitric oxide synthase) and iNOS (inducible nitric oxide synthase) expression of aorta, were investigated.
