單純庖疹病毒第一型 (Herpes simplex virus type 1; HSV-1) 是屬於庖疹病毒科 (Herpesviridae) 的病毒,根據血清流行病學的調查約有 90% 的人口都曾受過其感染。此病毒感染所造成的臨床症狀為在口部周圍造成潰爛現像,當其在個體造成初次感染 (Primary infection) 後,單純庖疹病毒第一型會於三叉神經結 ( Trigeminal ganglia) 形成潛伏期 (Latent phase),當個體接受過多紫外線照射、免疫反應降低或者內分泌反應失調,常會造成單純庖疹病毒第一型復發感染 (Recurrent infection),此種反覆復發的現像常造成個體莫大的困擾,也是現今社會普遍存在之慢性疾病之一。此外,庖疹病毒感染亦是愛滋病人中嚴重之醫療問題,現有之藥劑往往會抵消愛滋病治療效果或者造成毒素沈積骨骼之副作用,目前在臨床上並無有效治療方法,因此促使我們想由中草藥中找尋有療效但無毒性之中草藥。近年來,我們由一連串中草藥篩選工作中,發現冬蟲夏草 (Cordyceps sinensis) 子實體之甲醇抽取物能有效抑制單純庖疹病毒第一型的複製,由於有如此的初步結果,因此,在八十七年度衛生署研究計畫中,我們與陳建志老師合作,配合生物活性追蹤法進行有效成份分離,發現CS-F3-C6 具有很強之抑制活性,所以在本年度研究計畫中,我們將延續實驗進行機制之探討,實驗中我們將以Southern blot analysis 及Northern blot analysis 分析 CS-F3-C6 是否藉著抑制病毒 DNA 及RNA 而達到抑制單純庖疹病毒第一型複製的結果。

According seroepidemiology study, 90% of human population have been infected by herpes simplex virus type 1 (HSV-1). After primary infection, HSV-1 get into the trigeminal ganglia then set up a latent phase. When individual were in immunosuppressed, in much UV-light exposure or in disregulated endocrine secretion, HSV-1 will recovery from trigeminal ganglia, infect target tissues again and cause lesions around lips. The people are suffered from recurrent infection. So far, there is no efficient medicine to treatment this disease. Cordyceps sinensis is a major parasitic fungus on larvae of Lepidoptera. It is one of the well-known fungi used in traditional Chinese medicine. Many fungi belongs to the genus Cordyceps have been demonstrated to produce natural products with various biological actives.
