欖仁葉之抗遺傳毒性研究

由於近年來在台灣民間常用乾枯的欖仁樹落葉煎水做茶飲,或與茶葉混用沖泡開水服用,相傳有治療肝病的效果。但是其確切的療效及機轉目前並不清楚,所以有興趣對欖仁葉的用途做進一步的探討。另外在最近我們的研究發現,欖仁葉的水萃取物可以預防並? 磻螸itomycin C 所誘發的微小核(micronucleus)產生。因此計畫想朝向欖仁葉的抗遺傳毒性進行。
我們將先用由低到高各種濃度的欖仁葉水萃取物(Terminalia catappa extract, TCE)處理細胞,以確定對細胞無毒的最高劑量,再用細胞聚落形成,hgprt基因突變,及單細胞電泳方法進行對細胞及DNA保護效果的測試實驗,分析TCE可抑制哪幾類致突變劑的傷? `,並測量細胞內抗氧化解毒酵素的活性。希望可以確定欖仁葉的抗遺傳毒性效果及可能透過的機轉。
此外,用有毒劑量的TCE處理細胞,以細胞聚落的形成來確定TCE的毒性,並進一步分析其對細胞週期,DNA合成的影響,還測定是否會引發細胞凋亡(apoptasis)的現象。希望可以確定出服用欖仁茶的安全劑量範圍,以及過量服用可能造成的細胞傷害。

Studies on the Antigenotoxic Effect of the Leaf of Terminalia catappa.

The fallen leaves of Terminalia catappa L. have been commonly used for treating liver related disease in Taiwan, yet the definite effects and mechanisms remain elusive. In recent study showed that the water extract of Terminalia catappa L. (TCE) was able to prevent mitomycin C-induced micronuclei and Fe2+/H2O2 induced lipid peroxidation. So we are interested in examing the antigenotoxic effects of the herbal drug.
The cytotoxicity of TCE will be determined first by observing clonies formation. And the antigenotoxic effects of subtoxic dosage TCE will be detected by hgprt mutation and single cell gel electrophoresis assay. Then the antioxid ant enzymes activities will be measured. In addition, the cytotoxic effects of TCE will be studied. Cell cycle, DNA synthesis and apoptosis after toxic dosage TCE treatment will be determined.
