延胡索成分四氫巴馬汀對熱中風動物模式之療效評估

我們過去的研究報告指出熱中風發生時動物的動脈壓下降,單胺濃度昇高,顱內壓昇高,腦局部血流減少,神經細胞損傷以致于動物的存活期縮短至15分鐘左右。如果事先將動物腦中的單胺物質(尤其是妥巴胺或5- 色胺)濃度降低,則可以延長熱中風動物的存活期並減輕熱中風動物的低動脈壓、高顱內壓、腦缺血及細胞損傷等症狀。最近我們也發現中藥延胡索成分四氫巴馬汀具有很強的降低動物腦內單胺物質濃度的作用,因此我們擬在本實驗中探討四氫巴馬汀對於熱中風動物模式之治療效果。首先,我們建立熱中風動物模式。將全身麻醉的大鼠暴露置于高溫環境(42℃)以確定熱中風發生時出現低血壓、高顱內壓、腦缺血、細胞損傷及縮短存活率等現象。然後觀察當給予四氫巴馬汀治療後,上述熱中風症狀群是否被改善。從本實驗預期的結果可以得知延胡索成分四氫巴馬汀對於熱中風動物模式之療效有多大,且其經由何種機制來達其療效。本實驗之預期成果不僅可在熱中風生成的學理上有所建立,並且可提供熱中風臨床治療上的參考之依據。本計畫擬同時探討在熱中風下四氫巴馬汀對於全身動脈血壓、血流、血管阻力及體液電解質之影響。

Our previous results have shown that the animals with heatstroke displayed arterial hypotension, intracranial hypertension, cerebral ischemia, neuronal damage and shorted survival. Depleting monoamines (in particular the dopamine an d serotonin)in the brain with neurotoxin significantly extended the survival of the animals with heatstroke and attenuated the heatstroke-induced arterial hypotension, intracranial hypertension, cerebral ischemia, and neuronaldamage. Recently, our result s also demonstrated that the monoamine levels of rat brain were greatly decreased after systemic administration of dl-tetrahydroxy-palmatine. Therefore, the present study will assess the effects of systemic administration of dl-tetrahydroxypalmatine on t he survival of the heastroke animals and the heatstroke-induced arterial hypotension, intracranial hypertension, cerebral ischemia and neuronal damage.
