中草藥應用於保健食品開發之安全性研究

本計畫擬利用一年的時間篩選台灣名間鄉所栽種之生薑(Zingiber officinale Roscoe)、刺五加(Acanthopanax senticosus)及薏苡(Coix lachryma-jobi L.)三種藥材配方組合,應用於體重控制。首先測定藥材是否有重金屬和農藥殘留並檢測藥材指標成份含量。並以不同炮炙方法及溶媒萃取進行最佳配方比例篩選製造成膠囊劑型。本研究運用3T3-L1細胞針對研究中不同藥材比例配方探討脂肪細胞增生及分化率測定之影響,將待測中草藥膠囊劑型進行健康食品法所規範之安全性測試:急性毒性試驗、28天餵食毒性試驗及基因毒性試驗共三項,所得結果可為日後當為「不易形成體脂肪功能」之健康食品安全性劑量的參考。

Safety evaluation on the Chinese herbal medicine for health food development

The research is the two-year plane. The aim of the research is going to evaluate the effects of herbal medicines and the processed products that commonly used in weight reduction, on prevention of high energy diet-induced obesity in rats. In addition, the possible hepatotoxicity or nephrotoxicity induced by long-term administration of these anti-obesity herbal medicines and its processed products will be evaluated. In the first year, the anti-obesity efficacy and the toxicity associated with herbal with hypolipidemic activity, such as Polygoni Multiflori Radix and its black bean sauce-processed product, as well as Bupleuri Radix and the vinegar-processed product, will be evaluated. In the second year, the anti-obesity efficacy and the toxicity related with herbal diuretic, including Alismatis Rhizoma and salt-processed product, or Cassia Seed and firing-processed products, will be investigated. During the experimental period, the plasma levels of glutamic pyruvic transaminase and glutamic oxaloacetic transminase will be measured, and hydroxyproline content will be determined in liver tissues to evaluate whether the hepatotoxicity will be induced by the tested herbal medicines and the proceed products. Kidney damage induced by the tested herbal medicines and the processed products will be determined by chaning in the levels of blood urea nitrogen and plasma creatinine concentration. The biopsy will be further performed to evaluate the pathology of hepatotoxicity and nephrotoxicity. Furthermore, the influences of the tested herbal medicine on the levels of transforming growth factor-β1, a protein related with extracellular matrix synthesis, as well as the changes of collagen IV, will be determined by immunohistochemistry staining. The results will provide valuable information about safety of tranditional Chinese medicine for both academic and industrial parties and maybe helpful for the establishment of related policy.
