常用中藥對人類細胞色素P450影響之研究

由於民眾對傳統中草藥的普遍使用，中藥及天然物影響藥物代謝酶所造成的中西藥物交互作用的問題也日益受到重視。在細胞之內質網膜上，由細胞色素P450 (cytochrome P450，簡稱P450) 組成之單氧酶系統是主要負責外來物氧化代謝的酶系統，這些外來物包括：藥物及天然物。在這一年期計畫中，研究目的包括：(一)建立人類細胞色素P450之表現及活性分析系統；(二)常用中藥萃取物對人類細胞色素P450之體外作用；(三)代謝對常用中藥萃取物與人類細胞色素P450交互作用之影響。研究工作包括：(一)以大腸桿菌DH5脉，建立最佳之人類細胞色素P450表現及活性測定方法。(二)建立枸杞、大棗、茴香、延胡索、薏苡仁、山藥萃取物之層析圖圖譜，質譜圖譜。測定人類細胞色素P450之活性受中藥萃取物之影響。(三)分析比較中藥萃取物代謝前後之層析圖，質譜圖，以說明是否有P450代謝反應。比較有及無代謝反應後對P450抑制作用之影響，以了解代謝作用在交互作用中的角色並說明mechanism-based inhibition的機轉。進行這些研究對於現今社會，民眾用藥安全將有很大的幫助。
