利用分子模版之技術自中國傳統藥材提煉異黃酮素之防癌藥劑

本研究計畫的目的在於希望利用生物分子模版(MIP)的方法,代替傳統的分離純化程序,自中國食品及藥材中提煉異黃酮類物質,主要技術即利用所要提煉的異黃酮類物質製造出具有高辨識能力的高分子模版,以此具有選擇性的高分子模版從天然植物中提煉並純化異黃酮類物質,如此便可突破以往傳統分離純化步驟的費時及耗能的問題。近年來由於大豆本身內含的營養成分豐富,對於人體健康相當有益,並且在日常飲食中扮演重要的角色,因而備受重視,且由近期的普查結果顯示,國人對於蛋白質的攝取比起其他國家仍然不足,而大豆中含有非常豐富且容易被人體吸收的植物性蛋白質,可幫助身體結構進行修補,以及調整人體各機能的運作,強化肌肉組織,綜合上述大豆的優點逐漸的被各國所重視。美國將每年的八月十五日訂定為全國的〝豆腐節〞;中國大陸的衛生部提出了〝大豆行動計劃〞,各國對於大豆的重視由此可知。除了優質植物性蛋白質,大豆所含有的其他營養成分也不容忽視,其中大豆異黃酮類物質為近幾年許多研究人員積極研究的物質,大豆異黃酮類物質的結構式和雌激素非常相似,是一種具有雌激素活性的植物性雌激素,尤其在於醫療領域的應用上,由於大豆異黃酮類物質具有抗老化的作用,可以延遲細胞的衰老,使得皮膚保持彈性,以及具有能夠減輕女性更年期綜合症症狀的效果,促成骨質生成而減少骨質流失,並且可以有效地調節血脂,降低膽固醇保護心血管及抗癌等作用,已知大豆異黃酮類物質具有上述多種藥效,對於醫療以及藥物應用領域具有相當的貢獻。

Extraction of Genistein and Biochanin A From Chinese Traditional Herb by Molecularly Imprinted Polymers

Molecular imprinted technique was substantial succeeded in the field of biomolecular sensor and solid phase extraction. Molecularly imprinting polymers (MIPs) are polymeric materials possessing highly selective and affinitive properties. Flavonoids, which are widely distributed in natural fruits and vegetables, contribute positive effects to health, due to their anti-oxidant, anti-carcinogenic, anti-allergy, and anti-inflammatory activities. Recently, many researchers reported that isoflavones from Chinese traditional herb had the functions of antioxiation and used as anticancer drug trails. In this study, we will focus on the recognition and purification of two kinds of isoflavonoid deritives, genistein and biochanin A. Both of them belong to the isoflavone class of flavonoids, and they have estrogenic activity because their structures are similar to the structure of the estrogenic hormone. So, under conditions of law estrogen exposure, such as during menopause, the isoflavones tend to act as weak estrogens, which may be just enough to help alleviate some of the symptoms associated with menopause, such as hot flashes, headaches and mood swings.
