苦寒性中藥對於蛋白質酪胺酸激之作用

苦寒性中藥,傳統上是被用於實症狀態的病症,但其作用的機轉一直沒有被現代醫學所了解,我們綜合過去的研究,推測這類藥物可能是經由抑制蛋白質酪胺酸激 (tyrosine kinase) 的活性而完成作用,為了證明此一推論,我們將先建立一套篩選蛋白質tyrosine kinase抑制劑的方法,進而利用它來探討苦寒性中藥是否有此一作用。我們將以苦寒性中藥的萃取液,與經LPS或其他可促進tyrosine活化的物質刺激後的單核細胞作用,以決定該中藥是否有抑制作用,對於有作用之苦寒藥再進一步分離其有效成份及探討其作用機轉。

The effect of “Ku-Han” herbs on the protein-tyrosine kinase

The "Ku-Han" herbs have been used for treating patients with "Shu-Cheng" (實症), a symptom when the disease is in a strong reaction state, in Chinese medicine. However, the exact mechanism of these herbs reaction is still unknown. It has been hypothesized that these herbs may achieve the effects by inhibition of the protein kinase or protein kinase-related protein. The aim of this proposal is to study this inhibitory mechanism on the molecular level. A series method will be developed for screening the "Ku-Han" herbs to explore their inhibitory effect on protein kinase. Moreover, the effective components of these herbs will then be identified and the detail pathway of protein kinase inhibition will be described.
