中藥有效成份catechin對kainic acid誘發癲癇發作老鼠的抗癲癇作用機轉之研究

癲癇是自古以來已存在的疾病,醫學發達的今日,雖然許多抗癲癇的藥物的發現,但癲癇仍然被列為難治的疾病之一,尤其是複雜性部份性癲癇。在前年和去年我們的研究發現kainic acid誘發wet dog shaking的最大發作頻率是在kainic acid投予後的60和80分之間,中藥鉤藤的粗萃取物、天麻的有效成份vanillyl alcohol和中藥的有效成份catechin可以減少kainic acid誘發的wet dog shaking頻率。同時我們也發現鉤藤的抗癲癇作用和它的抗脂質過氧化作用有密切的關係。因此我們今年進一步的探討catechin的抗癲癇作用機轉。首先,將30隻重量200-250 g的雄性Sprague-Dawley(SD)老鼠分成5組,每組6隻。我們分別測定它們在kainic acid(12 mg/kg)腹腔注射前,注射後30分,60分,120分和180分時大腦皮質的脂質過氧化物濃度和末梢血液的luminol-chemiluminesenence(CL)及lucigenin-CL值。其次我們測定catechin於體外的抗脂質過氧化作用,使用SD老鼠的大腦皮質和kainic acid混合,然後加入不同濃度的catechin後分別測定它們的脂質過氧化物濃度,同時和carbamazepine及vitamin E做對照。最後﹐另外將18隻SD老鼠分成3組,每組6隻如下:A)catechin 100組:我們於kainic acid投予前的30分鐘分別腹腔注射catechin 100 mg/kg,然後於投予60分鐘後分別測定它們大腦皮質的脂質過氧化物濃度,末梢血液中的luminol-CL及Luciganin-CL和值;B)catechin 50組:方法同A,但使用catechin 50 mg/kg;C)carbmazepine組:方法同A,但使用carbamazepine 20 mg/kg。
根據我們的結果我們可以進一步了解中藥的有效成份catechin的抗癲癇作用機轉。

The anticonvulsant mechanisms of catechin component of Chinese Herbs in rats with kainic acid-induced epileptic seizure

Epilepsy resulted from abnormal synchronized discharge of neurons in brain has been seen since ancient time. Although modern medicine has good developments, the epilepsy, especially the complex partial seizure, is still considered one of the most intractable diseases. In our previous studies, we found that the maximal frequency of wet dog shakes happened between 60-80 min in rats after the administration of kainic acid (KA). However, it was found that uncaria rhynchophylla (Miq) Jack, vanillyl alcohol and catechin could significantly reduce the count of wet dog shakes. We also observed that the anti-convulsive effect of uncaria rhynchophylla was closely related to its suppressive effect on lipid peroxidation. In this study, we further study the anti-convulsive mechanisms of catechin. First, 30 Sprague-Dawley (SD) rats, weighing between 200-250 g, were divided into 5 groups of 6 rats.
