補益劑對高血糖鼠學習記憶能力之影響及機制探討

現已知初期癡呆症患者之臨床主要徵候為學習記憶之能力逐漸減退,特別是近期記憶喪失及剛學習之事物無法記得。另在臨床及動物研究上,均發現糖尿病患者或實驗動物給予streptozotocin可造成記憶能力及學習操作障礙之現象。但亦有研究,認為智能增強劑之增強學習記憶作用,可能經由影響腎上腺之機能,使體內之glucose使用率增加,腦內血醣濃度增加,因而增加腦內能量之供給,活化腦內神經系統;並指出glucose可增強智能及記憶操作能力。因此血糖之多寡應與學習記憶之能力有密切之關聯性。

  著者近幾年內針對中藥方劑在記憶增強作用所進行之篩選研究中,發現補氣類方劑補中益氣湯、補血類方劑歸脾湯、補陰類方劑天王補心丹及補腎類方劑六味地黃丸均可改善正常大鼠因藥物所造成之學習記憶障礙;另著者研究報告亦指出知柏八味丸具降血糖作用。因此,本研究擬將上述5組方劑進行水抽取後,以被動迴避學習反應來探討對正常大鼠因cycloheximide所誘發學習記憶障礙之預防或治療作用及對streptozotocin誘發高血糖鼠所誘發學習記憶障礙之預防或治療作用;另因在被動迴避儀之實驗中仍受大鼠運動活性及對電刺激引起的痛刺激反應影響,因此本研究另輔以自發運動量及電痛閾值之測定。其次,並進行最有效方劑水抽取物對正常大鼠及streptozotocin誘發高血糖鼠葡萄糖耐受性、學習前後血漿及胰臟中glucose、insulin及glucagon濃度之變化。

Studies on the action mechanism of the ameliorating effects of Pu-I prescriptions on memory dysfunction in streptozotocin-induced diabetics mellitus rats

The population of aging is gradually increasing, the patients suffered from dementia are simultaneously increasing. The mainly symptoms of the dementia involved the decline of cognition function, especially the decline of recent memory and easily forgotten for recent learning experience. The existing cognition activators possess various levels on the improving effects for various drugs-induced learning and memory impairment, but these drugs in clinical application did not develop their efficacy. In clinical and basic pharmacological reports, diabetic patients ensures the memory impairment and the streptozotocin-induced hyperglycemic rats also impaired acquisition and retention compared with normal rats.

 Our previous reports pointed out that some “Pu-I prescriptions” such as Pu-Chung-I-Chi-Tang, Gui-Pi-Tang, Tien-Wang-Pu-Hsin-Dang and Liu-Wei-Di-Huang-Wan improved the various drugs-induced learning and memory impairment. We also reported that Ba-Wei-Di-Huang-Wan had the hypoglyce
