中醫治腦方劑對腦血管老化、腦中風療效的實驗研究

腦血管老化會導致腦中風，而腦中風是國人第二大死因，更是老年人最常見的疾病之一。利用核磁共振攝影，可快速診斷是否有腦中風引起之腦組織缺血，甚至腦細胞老化引起腦梗塞。但在治療腦中風方面，西醫仍無有效藥物可供使用。中醫治腦方劑流傳已久，有些有臨床療效。本研究目的即在探討這些中醫治腦方劑，是否具保護缺血腦組織，使缺血腦細胞不產生老化，防止腦梗塞之產生及其可能之保護機轉。

本研究將採用大白鼠大腦皮質缺血動物模式來測試中藥方劑之療效。使用大白鼠重約450公克，利用chlorohydrate (400mg/kg)腹腔注射麻醉，利用顯微手術，可把雙側頸動脈及右側中大腦動脈結紮，90分鐘後再放開，使大腦皮質血液恢復流通。24小時後，大白鼠再次麻醉，取腦作2%的triphenyl tetrazollium chloride (TTC)染色，梗塞腦皮質區會呈白色。 

16種不同的治腦方劑將逐一測試其對缺血性腦損傷之療效。這些方劑將依臨床病患服用方式調理，然後灌食大白鼠三天，再行暫時性血管結紮手術，再測動物大腦皮質梗塞體積，即可知曉是否具療效。

我們及他人的研究也顯示在缺血腦組織內一氧化氮(NO)會大量的產生，並造成腦細胞之老化死亡。抑制一氧化氮之產生，即可減少腦梗塞。若中藥治腦方劑顯示具療效，我們則將利用一氧化氮電極測試是否中藥方劑也可減低一氧化氮之生成，以致減低腦梗塞。本研究是利用最新的科學性動物模式來測驗出具保護缺血性腦組織的中藥方劑，並進一步了解是否與抑制一氧化氮自由基有關。如此可使中醫方劑對治療腦中風療效具科學證據。進而可再分別測試單味中藥之療效及其有效物質之純化，發展治療腦中風之新藥物製作。

Therapeutic effects of Chinese medicine on the cerebrovascular aging and stroke

Atherosclerotic changes of cerebral vessels usually result in cerebral infarction which is the second leading cause of death in our country. It is also a common disease in elderly people. By using MRI, a quick diagnosis of either ischemic brain damage or neurodegenerative disease can be made. Although, in western medicine, there is no effective treatment of cerebral infarction at the moment, some Chinese medicine is reported to have some kinds of effectiveness in stroke treatment. The aim of this study is to examine whether some prescriptions of Chinese medicine can (1) protect brain against ischemic damage, (2) prevent neurons from degeneration and (3) reduce the incidence of cerebral infarction. The mechanism of the protection is to be studied as well.
