利用定量聚合酉每 鏈反應評估抗老化方劑對於單核球細胞及纖維母細胞超氧化物歧化酉每 的影響

老化是自古以來困擾人類的課題之一,在邁向老年化社會的今天更形重要。抗老化方劑或補益方劑是傳統中醫藥中特有的一環,但其療效缺乏科學的驗證。老化的成因眾多,其中自由基學說最為吾人所接受。本研究即針對中藥能否增加人類單核球細胞及纖維母細胞超氧化物歧化酉每 之活化,用分子醫學的模式,以定量聚合酉每 鏈反應直接測定各細胞超氧化物歧化酉每 的傳訊核醣核酸,其優點為其偵測範圍度及靈敏度高且容易對中醫藥的治病機轉提出強而有力的證據。藉此技術,吾人可大量篩檢傳統中醫藥中之抗老藥方,並探討其臨床意義。

The effect of Chinese anti-aging medicine on superoxide dismutase evaluated by quantitive PCR

The anti-aging effects of two Chinese medicines were evaluated in cultured human embryo lung diploid cells. In order to search for a possible mechanism of aging, the enzyme activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were measured in different generations of cultured cells. Paraquat was introduced into some groups of culture cells to generate free radicals and produce cell damage. The enzyme activity of SOD and GSH-Px increased remarkably in the groups treated with Luh-Wey-Dih-Hwang-Tang (六味地黃湯). The life span of cells in this group was significantly prolonged when compared with the control group. The anti-aging effect of Luh-Wey-Dih-Hwang-Tang may come from the activity enhanament of SOD and GSH-Px. The life span was shortened in all groups of cultured cells treated with paraquat. The group treated with paraquat and Syh-Jiun-Tzyy-Tang (四君子湯) had longer life span than the other groups treated with paraquat. The anti-aging effect of Syh-Jiun Tzy
