紫雲膏中抗炎成份naphthoquinones類對中性白血球NADPH oxidase活性的影響

中性白血球為一種重要的發炎細胞,在保護個體防止病菌侵害上扮演重要的角色。當受到刺激會產生超氧自由基其他有毒的含氧代謝產物來殺死侵入的病菌,但過多的有毒含氧代謝產物,也會傷害到周圍的正常組織。已知有磷酯鋂丙、蛋白質激鋂丙、磷酯鋂丁、酪氨酸激鋂、以及磷酸醯肌醇三激鋂活性的增加等細胞內的訊息傳遞與中性白血球的NADPH氧化鋂活化有關。在氧化鋂活化的過程裏,有細胞質中的p47phox、p67phox及rac會轉移至細胞膜上與cytochrome b558結合,行成一個具有活性的NADPH氧化鋂複合體。在NADPH的存在下,可對氧分子進行單一電子的還原產生超氧自由基。紫雲膏用於治療燒傷、凍瘡及痔瘡等疾病,可促進組織肉芽的生長及防止皮膚潰爛。紫雲膏中的紫草具抗炎和促進創傷愈合的作用。紫草中含naphthoquinones類的抗炎成份其中acetylshikonin已知具有抗皮膚血管通透性增大、抑制浮腫、及抑制中性白血球生成超氧自由基的作用,為紫雲膏療效的主要成份。Acetylshikonin僅略覣微抑制磷酯鋂丙及磷酯鋂丁的活性,但有強的抑制蛋白質酪氨酸磷酸化的增加的作用。本計畫擬比較紫雲膏與acetylshikonin對中性白血球呼吸爆發作用影響的差異,探討紫雲膏中acetylshikonin含量變化和安定性,並進一步探討acetylshikonin是否會影響細胞內磷酸醯肌醇三激鋂的活性,及對NADPH氧化鋂複合體形成的影響。經由這些實驗將更能了解紫雲膏的療效成份acetylshikonin對中性白血球生成超氧自由基的調控機制。

Effect of naphthoquinones, the anti-inflammatory ingredients of tzu- yun-kao, on the NADPH oxidase activity of neutrophils

Neutrophil is a prominent inflammatory cells, plays a critical role in host defense against infection. Upon stimulation, neutrophils release large amounts of superoxide anion and other toxic O2 metabolites. They are believed to serve as bactericidal agents. However, the excessive release of these reactive O2 species can result in undesirable tissue damage. It is known that the activation of PLC, PKC, PLD, TK and PI3K signalling pathways are involved in the activation of NADPH oxidase, in which the assembly of cytosolic factors (p47phox, p67phox) to the membrane cytochrome b558 occur and then produce superoxide anion, in the presence of NADPH, through univalent reduction of O2. Tzu-yun-kao is a Chinese medicine for the treatment of burn, chilblain and hemorrhoid, promoting granulation of tissue and preventing dermal erosion.
