川芎口秦對氣喘動物模式治療之探討

氣喘是以氣道變應性炎症,支氣管過度反應性為特徵的疾病,治療上強調抗炎為主的綜合治療,近年來研究得知,控制支氣管過度反應性和難治性哮喘與氣管變應性炎症,氣道上皮損傷,血液高凝狀態,血小板活化因子,鈣通道阻滯劑之間有密切關係,氣喘發作時伴有凝血功能亢進,其中血小板活化因子作用很受重視。血小板活化因子不僅激活血小板功能,而且是引起氣喘的重要介質,凝血功能亢進與血小板活化和血小板活化因子的關係,提示中醫活血化瘀法是治療氣喘重要的方法,事實上臨床運用活血化瘀方藥治療難治性氣喘亦獲得較好的效果,其中川芎口秦是最常用的藥物,川芎口秦能抑制血小板的聚集和釋放,又具鈣離子拮抗作用及免疫調節功能,其對氣喘之防治應有一定作用,惟探討之文獻不多。

本研究擬利用塵純化液誘發天竺鼠形成氣喘動物模式,藉身體體積量法、肺泡沖洗液分析、分子生物學、肺部病理組織檢查等方法,研究氣喘天竺鼠,以川芎口秦治療後其呼吸道阻力、呼吸道發炎細胞、發炎介質、免疫功能之改變,以了解川芎口秦對氣喘動物呼吸道阻力、呼吸道發炎細胞與肺部病理組織之影響,期望能了解川芎口秦對防治氣喘之影響。

The Study of Tetramethylpyrazine on Experimental Animal Model

There is close relationship between bronchial asthma and bronchial hyperresponseveness airway inflammation bronchial epithelial injury hypercoagulation platelet activating factor Ca+2 antagonist. Hypercoagulation after occur in acute asthma attack, ”Quicken the blood and transform stasis” is therapentic method in treating bronchial asthma in chinese medicine. Tetramethylpyrazine, is a biologically active component originally a popular chinese nerbal medicine Rhizoma liqustici wallichi, will be useful in asthmatic treatment due to its biological activities including antipleldet aggregation inhibition of both Ca+2 influx and intracellular Ca+2 release and immune modulation.

In this research, we use mite to induce guinea pig to establish asthmatic animal model. We evaluate the change of respiratory resistance and inflammatory cell of respiratory tract of sensitized guinea pig before and after tetramethylpyrazine treatment by body-plethysmography, bronchoalveolar lavage and bronch
