四君子湯活體內調解免疫球蛋白製造的作用機制:第十介白質的角色

四君子湯為補氣健脾的基本處方,含黨參、白朮、茯苓及甘草。本實驗室歷年來之研究結果顯示,四君子湯對老鼠及人類IgA之分泌,皆有顯著的促進作用。這種效應與四君子湯中的茯苓有關。誘導B淋巴球製造IgA的主要細胞激素之一是轉型生長素(TGF-β)。本實驗室最近研究茯苓對TGF-β製造的影響,結果顯示,茯苓可抑制TGF-β的製造。此一結果與先前促進IgA製造的發現不符,故本計畫研究目標為探討另一誘導B淋巴球製造IgA的主要細胞激素—第十界白質(IL-10)在四君子湯調節體內免疫球蛋白製造時的角色。老鼠將腹腔注射或口服四君子湯及茯苓之沸水萃取液,連續三天後取出血液,以ELISA測血清中IgA、IgG、IgM及IL-10濃度,在此同時取出脾臟,分離脾臟淋巴球,離體培養24h,測其IL-10分泌能力;培養4天後,測其IgA、IgG及IgM的分泌能力。研究之最後階段將以抗IL-10抗體中和老鼠體內之IL-10,探討IL-10對四君子湯及茯苓在活體內藥效的影響。本研究將有助於吾人進一步瞭解四君子湯在活體內調節免疫功能的作用機轉。

The mechanism that mediates the regulatory effect of Si-Jun-Zi-Tang on immunoglobulin production in vivo: the role of interleukin-10

Si-Jun-Zi-Tang is one of the widely used Chinese herbal medicines. Si-Jun-Zi-Tang (SJZ Tang) consists of four major ingredients, Dongshen, Baizhu, Gancao and Fuling. Our previous studies demonstrated that SJZ Tang significantly enhanced immunoglobulin A (IgA) production in both murine and human system. This biological activity might be derived from Fuling since Fuling was the only ingredient that showed similar effect on IgA production. However, our recent report suggested that Fuling suppressed the production of transforming growth factor-b (TGF-β), One of the cytokines that induced IgA production by B lymphocytes. This observation apparently conflicted our previous data that Fuling played a position role in IgA production. Therefore, the specific aim of the present project is to investigate the role of interleukin-10 in SJZ Tang-mediated immunoglobulin production. Mice are i.p. injected or orally adminstrated with ot-water extract of SJZ Tang and Fuling for three consecutive days.
