知母指標成分之製備

知母為一常用中藥，漢方以消除煩熱、口渴等為目標時可作為解熱、消炎、鎮痛、利尿之用。應用的製劑有桂芍知母湯、酸棗仁湯、滋陰降火湯、白虎湯等。有關化學組成，主要包含了Sapogenins，Saponins，Norlignans及Xanthones類化合物；特別是以Sarsasapogenin，Timosaponin A-III及Mangiferin為主。藥理實驗證實，知母中的Saponins及Sarsasapogenin的抗熱作用；亦証實Mangiferin具有抗炎、抗結核、利膽及中樞神經抑制作用等活性。本計畫擬從中藥知母中，分離純化指標成分Sarsasapogenin 、Timosaponin A-III和 Mangiferin；預定分得上述純指標成分Sarsasapogenin (1 g)、 Timosaponin A-III (1 g)、Mangiferin (5公克)，並完全提供衛生署貴會使用，並建立其純度規格及HPLC的分析方法，以提供QC部門的參考應用。

Preparation of the Maker Compounds of Rhizoma Anemarrhenae

Zhimu,, Rhizoma Anemarrhenae, is the dry rootstock of Anemarrhena asphodeloides Bge. (Liliaceae) drug in spring and fall. It has been used for a long time in traditional Chinese medicine as an antipyretic, sedative and diuretic agent.

The main constituents of the rhizome of A. asphodeloides are saponins, spogenins, norlignans and xanthones etc.. Especially, sarsasapogenin, timosaponin A-III and mangifernin are the representative substances of A. asphodeloides. The saponin fraction isolated from the rootstock of A. asphodeloides and its hydrolysis product, sarsasapogenin showed potent inhibitory action on Na+/K+ ATPase which the activity may be related to its antipyretic action. Mangiferin also exhibited antipyretic, sedative activities.
