藉由組織培養技術復育繁殖雷公藤

雷公藤是一目前用來治療風濕性關節炎、自體免疫疾病(如紅斑性狼瘡)的藥用植物。含有雷公藤素(trypterygine)、wilforidine、wilfodeine及wilforgine等生物鹼,具有抗發炎的作用,在大陸地區已研發成藥劑,國內則仍在開發階段。但由於國內環境之變化,造成雷公藤已濱臨絕種。有鑑於此,本計畫乃針對雷公藤以組織培養技術來進行繁殖復育,使研究材料不虞匱乏,除了避免絕種外,亦可使藥用品質穩定,經由癒合組織 (callus) 的萃取,更可使雷公藤素等生物鹼量提高。

由於組織培養技術的建立需要不斷的嘗試以得到最佳生長條件,故擬以二年來進行完整之實驗:
第一年:雷公藤組織培養技術之建立。
第二年:雷公藤組織培養後癒合組織的藥性分析與復育繁殖。
本計畫擬先建立雷公藤組織培養技術,以得到培養大量癒合組織之條件,提供將來藥性分析與復育繁殖之用。

To reproduce Tripterygium wilfordii Hook by the technique of plant tissue culturre

Thunder God Vine (Tripterygium wilfordii Hook; Celastraceae), a poisonous medicinal plant, has been clinically used for the treatment of rheumatoid arthritis and autoimmune diseases. It contains alkaloids (trypterygine, wilforidine.wilfodeine, and wilforgine). multiglycosides. triterpenoids and nontriterpenoids, possessing marked anti-inflammatory, analgesic and immunosuppresive actions, and has been well developed as pharmaceutical preparations for clinical use in China mainland. However, this plant is becoming extinct in Taiwan due to environmental changes. The aim of the present study protocol attempts to breed this plant by the tissue-culture technique and thus to prevent it from extinction. In addition, the main constituents in callus will be extracted and analyzed chromatographically.

Two years of researching period will be needed for the establishment of optimized conditions for the culture and the analytical method, the study protocol will be carried out within following
