三七所含之三亞油酸甘油脂之定量分析及動物體中之動力學研究

三七是一種常用的活血化瘀中藥，過去皆以皂做為其主要的有效成分。近年來我們發現三七含有一種「三亞油酸甘油脂」的有效成分，有改善紅血球變形能力，抑制血小板凝集，促進血管壁釋放血管放鬆因子之作用。由於此種成分過去未曾有藥劑之研究，故本計畫擬開發此成分之分離、精製及分析之方法，以便將此成分做為一種活血化瘀之新製劑。

本研究之第一部份係以高效液相層析研究在各種不同萃取溶媒中三亞油酸甘油脂之含量。第二部份則對靜注三亞油酸甘油脂之大白鼠及家兔投藥後定時抽血，再測定血中三亞油酸甘油脂之濃度。此種測定方法之回收率、準確度及精確度皆將確定。第三部份則以電腦分析血中三亞油酸甘油脂濃度，而研究其藥物動力學參數。同時並特別分析三亞油酸甘油脂在腦部之濃度，探討其在腦部的分佈，做為一種『組織濃度』的研究。

Quantitative analysis and Animal Pharmacokinetic Study of Trilinolein isolated from Panax notoginseng

Panax pseudoginseng is a frequently used Chinese medicinal herb. Most of the previous studies were focused on its saponin fraction. We recently purified from it an active component that improved erythrocyte deformability, inhibited platelet aggregation and induced the release of endothelium-dependent vasodilation. Pharmaceutical research of this active component, trilinolein, is lacking. In the proposed project, we will develop the methods for purification and identification of this component so it can be developed into a new therapeutic agent in the future.

In the first part of this project, we will measure the concentration of trilinolein in different extraction solvents with a high performance liquid chromatograph. In the second part, we will draw blood from rabbit and rat which have received intravenous injection of trilinolein. The serum concentration of trilinolein will then be measured. In the third part, we will determine the pharmacokinetic parameters of trilinolein by
