探討不同賦型劑對附子、細辛、杏仁、桂枝等 四種中藥材指標成分含量之影響

本計畫探討兩種中藥方劑麻黃附子細辛湯和麻黃湯中之四種中藥藥材(附子,細辛,杏仁,桂枝)之交互作用及其對四種常用賦形劑(乳糖,澱粉,磷酸氫鈣和微晶纖維素)之效應。

首先以適當的溶媒與方法抽取這四種中藥材料然後濃縮乾燥得乾粉。然後分別與四種賦形劑以1:1(W/W)的比率混合,所得的粉末分別置於兩種相對濕度(0%;75%)及三種溫度(35℃; 45℃;60℃)所構成的六種環境中儲存,並定時取出以高效率液相層析儀(HPLC)測定Aconitine(附子標品),Amygdaline(杏仁標品),Cinnamic acid(桂枝標品),Methyleugenol(細辛標品)之含量變化情形,並以阿倫尼亞斯方程式評估其架儲期。

藥物與賦形劑間之交互作用實驗是採單一之主成份與賦形劑混合(1:1),稱取約5mg置於鋁盤(Alumium Pan)中,蓋上鋁蓋密封後,於室溫下儲存一個月。以示差掃描熱分析儀(DSC) 來探討藥物主成份與賦形劑可能產生之交互作用。

本計畫預期利用HPLC及DSC所得之分析資料為濃縮中藥製劑中稀釋劑的選取找尋─較快速而可靠的篩選方法。

Studies on the Effects of Various Excipients on the Indicative Components of Four Chinese Herbal Medicines


In the present investigation four herbal medicines (Aconitum carmichaeli Debx., Prunus armeniaca L., Asarum sieboldi Miq., and Cinnamomum cassia Bl.) from two traditional chinese medicine formulae (Shi-Shin Tang and Ma-Huang Tang) were chosen. Mainly the stability and possible interaction of the constituents of these four herbal medicines with four respective excipients (lactose, corn starch, dicalcium phosphate dihydrate and microcrystalline cellulose) would be studied.

The herbs were chopped and milled into fine pieces and extracted with suitable solvents, the extracts would then be freeze dried. Each freeze dried powder would be mixed(1:1 ratio) with the four excipients respectively, then tablets of these mixtures were prepared. The subsequent tablets would be stored in six different conditions and the storage effects on the constituents were monitored using a HPLC method periodically.
