雷公藤(tripterygium wilfordii hook. F., TWHF)為治療免疫疾患及炎症的傳統中藥材之一,其中所含活性成份triptolide是由雷公藤根部萃取之主要活性成分,已在臨床應用以治療如類風濕性關節炎等免疫缺失或發炎等疾病。近年的研究顯示triptolide同時還具有抑制腫瘤生長、腫瘤轉移及血管增生等功能;當合併與化療藥物處理時,可降低腫瘤細胞對化療藥物的抗藥性。胃癌為目前全球第二大致死癌症之一,好發於日本、南美洲、東歐國家及中東地區,臨床治療胃癌以外科、化學療法或兩者的合併治療為主,然而,療效卻因胃癌的高度轉移能力而受限。因此,發展有效控制胃癌細胞增生並抑制腫瘤細胞轉移之治療策略,為目前胃癌研究的重要課題之一。順鉑(cisplatin)是臨床上已經施用多年的化療藥物,因干擾細胞的核酸合成而誘發癌細胞走向凋亡,也常用於胃癌患者的治療,其所造成的正常組織破壞則限制了使用的劑量。Triptolide因具有免疫抑制及抗腫瘤細胞增生之作用,曾被認為可在與某些化療藥物合併時,強化抑制腫瘤生長的療效。然而,卻鮮少有triptolide加強化療藥物治療腫瘤的研究報告,為嘗試開發可強化化療藥物對腫瘤治療效果的中草藥,本研究乃以triptolide合併化療藥物進行對胃癌細胞影響之活體內與活體外評估及調控機制之探討。而triptolide與化療藥物併用時所產生的交互作用,亦會透過對胃癌細胞或將其植入活體誘發腫瘤生成後所進行的抗癌活性及抗血管新生研究進行深入的探討。

Effect of triptolide on cisplatin-induced growth inhibition and angiogenesis on tumor (2-2)

Tripterygium wilfordii hook. F. (TWHF), has been used as a Chinese medicine to treat patients suffering from immune diseases and inflammation for centuries. Triptolide, one of the active components extracted from the root of TWHF, has been used for the treatment of a variety of autoimmune and inflammatory diseases including rheumatoid arthritis. In addition to anti-inflammatory and anticancer activities, recent explorations of triptolide revealed many properties relevent to its inhibitory effects on tumor growth, metastasis and angiogenesis. It was reported to reduce the drug-resistance of tumor cells by combining with chemotherapeutic drugs. Gastric cancer is the second leading cause of cancer related deaths in the world. The highest incidence is found in Japan, South America, Eastern Europe and parts of the Middle East. Surgery and chemotherapy treated alone or combination are principal clinical strategies for gastric cancer. Till now, it remains a difficult disease to treat, primarily because of its aggressiveness and persistence. The development of new therapeutic and preventive strategies targeted at growth inhibition and metastasis of tumor cells could be an important goal to control gastric cancer cells. Cisplatin has been used clinically for several years on treating cancers by inducing apoptosis in cancer cells. According to the disturbing of nucleic acid synthesis and the subsequent progress of apoptosis, cisplatin was often used to treat patients with gastric cancer. However, there is rare research concerning about triptolide-enhanced therapeutic effect of cisplatin on cancer. For trying to develop traditional drugs on enhancing tumor therapeutic effect of chemotherapeutic drugs, triptolide is used to combine with cisplatin to evaluate the anti-cancer effect and regulatory mechanism of gastric cancer cells in vitro and in vivo. The interaction of triptolide and cisplatin is further investigated through the anti-cancer activities by both gastric cancer cells and tumor-bearing animal models in this study.
