常用中藥(附子、烏頭、天雄、半夏、大黃)與腎臟功能相關性研究

本計劃預計利用一年半的時間,針對目前市面上常用中藥,附子、烏頭、天雄、半夏及大黃等,利用動物實驗模式評估藥材可能引致之腎毒性。首先將針對一般市售產品進行資料調查,並利用藥材外觀及TLC進行中藥材的基原鑑定。之後將待測藥材以胃管給藥的方式投與至實驗大鼠。藉由實驗大鼠的血尿素氮(blood urea nitrogen)及血肌酐酸(creatinine )濃度的變化以及離體腎臟的組織病理切片評估實驗動物腎細胞腫脹及傷害的程度。同時探討腎臟中具腎功能調節作用因子的基因表現,以解明待測藥材引致腎毒性的可能病理機轉。本研究計畫的進行可為中藥安全使用提供重要依據,可提供產官學各界有關中草藥服用安全評估、政策方向及研究的參考。

Reasch on the functions of kidney influenced by the commonly used Chinese herbs-fuzi, wutou, tianxiong, banxia and dahuang

In an attempt to clarify the role of commonly Chinese herbals in the nephrotoxicity, fuzi, wutou, tianxiong, banxia and dahuang will be investigated in this proposal with 1 and half years experimental duration. First, he botanical of the tested commercial herbs will be evaluated by observation and TLC. Then, the rats will be fed with the tested herbs to ebaluate the levels of urea nitrogen and creatinine in blood. In addition, the kidney damage will be further determined by biopsy. In order to evaluate the pathology of nephrotoxicity induced by the tested herbs, the genes expression of regulators related with the functions of kidney will be further investaged. The results will provide valuable information about safety of Chinese herbals for both academic and industrial parties and maybe helpful for the establishment of related policy.
