魚腥草對抗單純皰疹病毒活性之機制探討


病毒感染長久以來都是人類健康與公共衛生上極欲克服的難題。臨床上,許多病毒感染造成的疾病都缺乏有效的藥物,例如困擾國人已久的肝炎病毒,至今仍無藥物可有效的對抗肝炎病毒。另外,2003年由冠狀病毒所引起之嚴重急性呼吸道症候群(SARS),在短時間內奪走多人性命,有鑑於此,抗病毒藥物得研發確實是一個重要的議題。
單純皰疹病毒(Herpes simplex virus)是一種普遍潛伏在人體的病毒,多數人都感染過此種病毒。單純皰疹病毒感染可引起嚴重的腦炎或全身性感染等症狀,嚴重者甚至導致死亡。受單純皰疹感染後,病毒會潛伏於體內,伺機復發。近年來,單純皰疹病毒感染的發生率與嚴重性皆有上升的趨勢,此與免疫功能有缺失的病人數目不斷增加有關。現形治療單純皰疹病毒感染之藥物acyclovir,使用量不斷增加,因而產生具有抗藥性之病毒,故急需發展新的藥物已治療單純皰疹病毒之感染。
魚腥草為一天然中草藥,已有文獻指出魚腥草對於某些病毒感染(如流感病毒)具有抑制效果。本實驗室於2001年也曾利用魚腥草測試其抑制單純皰疹病毒第一型之效果。實驗結果已初步證實魚腥草對於第一型單純皰疹病毒的感染具有抑制效果,然而先前的研究並未進一步分離魚腥草所含有效抑制病毒成分及進行其抑制病毒機制之探討。目前全世界也缺乏對魚腥草抗病毒成分及其作用機轉之研究。在此計畫中,我們期望測試魚腥草是否對第二型單純皰疹病毒感染也具有抑制作用,並進一步探討其抑制單純皰疹病毒之機轉。主要將利用高壓液相層析儀(High pressure liquid chromatography, HPLC)的方式分離出魚腥草中可能的抗病毒分子,並進一步利用核磁共振光譜(nuclear magnetic resonance s pectrometer, NMR)與質譜儀(mass spectrometer, MS)研究此抗病毒分子化學結構及分子量。最後希望能藉由微陣列晶片(Microarray)來瞭解魚腥草作用後對於細胞基因表現的影響

Study the mechanism of anti-Herpes Simplex Virus activity of Houttuynia cordata

Viral infection has been a long-term problem in human health and public health. Clinically, many diseases caused by viral infection lacked effective drugs. For example, hepatitis virus has long been plagued Chinese for a long time, but no drug can be effective against the virus today. In addition, coronavirus caused severe acute respiratory syndrome (SARS) in 2003, and caused more deaths in a short period of time, therefore, the development of antiviral drugs is indeed an urgent issue. Herpes simplex virus (HSV) is a common latent virus in humans, most people have been infected with HSV virus. HSV infection may cause encephalitis or serious systemic symptoms, and even cause death. HSV would lurk in the body, waiting recurrence. In recent years, the incidence and severity of HSV infection show upward trend, because the number of immuno-compromised patients increase continuously. The drug, acyclovir, for treatment of HSV infection was used increasingly currently and that caused the virus resistant to the drug. It is urgent necessary to develop new drugs for the treatment of HSV infection. Houttuynia cordata is a natural Chinese herbal medicine. Some study has pointed out the inhibitory effect of Houttuynia cordata for viral infection (such as influenza virus). In 2001, our laboratory had tested the inhibitory effect of Houttuynia cordata for HSV type 1.
