誘導枇杷癒傷組織之活性成分研究

枇杷為薔薇科植物Eriobotrya japonica Lindl.的乾燥葉,常用於止咳,清肺和胃,降氣化痰。其所含的主要成分有揮發油、三萜酸類、倍半萜類、黃酮類、多酚類、有機酸類等,具有抗炎止咳、降血脂、降血糖、抗病毒和抗腫瘤等活性。而其藥理、藥效的研究主要集中於三萜酸類成分(Tormentic acid , Maslinic acid , Corsolic acid, Oleanolic acid 和 Ursolic acid)。因此本計畫主要針對三萜酸類的活性成分利用植物組織培養方法,進行癒傷組織的誘導生產二次代謝產物。目前初步研究得知栽種枇杷葉,且容易受生長地區及季節變化影響其成分,採收又有一定季節限制,此外在萃取有效成分的步驟中,又受到大量葉綠素的干擾不易排除。因此若能利用組織培養獲得癒傷組織大量生產二次代謝產物,就能比自然植株生長快速且不受氣候之影響,也能大量減少萃取有效成分時所耗費大量的溶劑與時間。為利用枇杷資源作為三萜酸類成分的發酵培養以及工業生產和各種藥理活性利用上提供產業上的價值。

Eriobotrya japonica Lindl is a rosaceous evergreen tree distributed in southeastern China and Taiwan and has been used in traditional Chinese medicine for clearing away heat in the lung to stop cough and lowering the adverse flow of gi to stop vomiting. The leaf of the five main bioactive constituents were characterized spectroscopically as Tomentic acid, Maslinic acid, Corsolic acid, Oleanolic acid and Ursolic acid. The triterpenes compounds which show many biological and pharmacological activities, including anti-oxidative, anti-inflammatory, hepato-protective effects, anti-tumor, and anti-diabetes effects. in the study, we establish callus culture and analyze the callus for triterpenes by high-performance liquid chromatography . In the present study, we evaluated leaf of E. japonica from different areas in Taiwan and the contents of triterpenes will be affected by some reasons, such as the growing area and the differences between seasons. Therefore, there are lots of limit to harvest. In addition, its tough to avoid the interference of a great deal of chlorophyll in the process during extracting the active ingredient. Cell culture provides a method for the production of secondary metabolites and it will produce large amoumts of active compound and would not be affected by the climate and area. Also, saving the organic solvent and time. It provides the reference value of taking active compounds of E. japonica
