以動物實驗評估丹參及丹參酮ⅡA的作用

惡性腫瘤是台灣十大死亡原因的首位。丹參酮ⅡA是丹參(danshen)的提取物,有抑制癌細胞的增殖和引起凋亡的功效,我們先前研究證實丹参酮ⅡA濃度為1 μg/ml,colo 205細胞增殖作用明顯受到抑制,為對照組的56.54%。研究結果發現丹參酮ⅡA(2.5, 5 μg/ml)作用後,可引起colo 205細胞的p53、p21、fas、Caspase-8、Caspase-3和Cytochrome-c蛋白表現量上升。也發現丹參酮ⅡA,使細胞週期停滯在G0/G1期,引起人類colo 205細胞凋亡,抑制人類colo 205細胞的增殖。丹參酮ⅡA(2.5, 5 μg/ml)作用後,可引起colo 205細胞的NF-κB p65和MMP-2蛋白表現量下降,可以減少colo 205細胞侵襲和轉移能力。我們先前研究也證實丹参酮ⅡA對肺癌細胞株(A 549,H-292 ,H-661)有顯著的抑制作用,丹参和丹参酮ⅡA是有潛力的抗癌中藥,所以在本研究將進行體內實驗,來評估丹參及丹參酮IIA對肝臟是否有毒性反應;檢測丹参及丹參酮IIA影響各種肝功能指標;建立丹参及丹參酮IIA對肝臟之安全劑量,以作為日後臨床試驗的參考。

To Evaluate the effects of Dansen and Tanshinone IIA in vivo (2-2)

Malignant tumors were in the first place of the causes of death in Taiwan. Tanshinone II A was isolated from Danshen (Salviae Miltiorrhizae Radix), inhibited proliferation and induced apoptosis in human cancer cell lines. In our previous studies (in vitro) showed that the growth inhibition and apoptosis induction of tanshinone IIA on colo205 cell in a dose -dependent manner were obviously exhibited. Tanshinone IIA increase the levels of p53 , p21, fas , cyto-c, caspase-8 and caspase -3 in colo 205 cells by Western blotting methods, and through up- regulated the expressions of p53 and p21 then induced G0/G1 arrest and apoptosis. We also observed that Tanshinone II A affect levels of NF-κB p65 , MMP-2 and VEGF levels based on the Western blotting assays. Remarkably, Tanshinone II A not only exerted its effect on protein levels of NF-κB p65, MMP-2 and VEGF, but also directly inhibited the cell migration in vitro significantly was assayed by Boyden chamber methods. In our previous studies (in vitro) also showed that the growth inhibition and apoptosis induction of tanshinone IIA on human cancer cell lines in a dose -dependent manner were obviously exhibited. Danshen and tanshinone IIA are the potential anticancer traditional Chinese medicine, so we will to evaluate the hepatic toxicity for Danshen and tanshinone IIA by in vivo study, in order to be regarded as the reference of the clinical testing in the future.
