中西藥合併用於改善糖尿病腎臟病變之藥效及毒性評估

本計劃將針對中西藥合併用於改善糖尿病腎臟病變之藥效及毒性進行評估。將以「五苓散」以及西藥已知具有腎臟保護作用的Irbesartan，以個別投藥或是合併用藥的方式投予至由streptozotocin 所誘導的第1型糖尿病大鼠體內，藉由實驗大鼠的血尿素氮(blood urea nitrogen)、血肌酐酸(creatinine)濃度的變化以及離體腎臟的組織病理切片評估實驗動物腎細胞腫脹及傷害的程度。同時探討腎臟中具腎功能調節作用的因子如TGF-β1的基因表現，以解明中西藥合併用於改善糖尿病腎臟病變之可能病理機轉。並確認在此處方中何者是產生效果的君藥。本研究計畫的進行可為了解中西藥併用之藥效，提供產官學各界有關中藥服用及中西藥合併服用安全評估、政策及研究方向等參考。

Evaluation of the efficacy and the associated toxicity induced by Western medicine and traditional Chinese medicine combination on the diabetic nephropathy

In an attempt to clarify the influence of tranditional Chinese medicine on the development of diabetic nephropathy, Wulingsan, the trandition Chinese prescription, and Irbesartan, the angiotensin II receptor antagonist, as well as the two drugs combination will be given into streptozotocin (STZ)-induced diabetic rats. Changing on the levels of urea nitrogen and plasma creatinine concentration will be investigated. In addition, the kidney damage will be further determined by biopsy. In order to evaluate the pathology of nephrotoxicity, the genes expression of regulators related with the functions of kidney will be further investaged. Furthermore, the study will be clarify the imperial herbs of Wulingsan for the regulation on diabetic nephropathy.The results will provide valuable information about safety of tranditional Chinese medicine for both academic and industrial parties and maybe helpful for the establishment of related policy.
