建立枇杷懸浮細胞培養之研究

本計畫以開發本土中草藥植物資源永續使用為目的,利用枇杷細胞株建立懸浮培養方法並配合監控培養液成分變化,作為將來調控生物反應器大量生產有用物質之基礎研究。因固態癒傷組織的培養中不利於同調分裂,也無法於培養時控制營養液的比例而且培養空間受限導致無法同時大量表現有效成分。因此在之前研究枇杷癒傷組織固態培養之最好調控條件的基礎下,計畫進行液態培養探討大量生產細胞及有效成分的可行性。
台灣本土枇杷來源不虞匱乏,可以每年誘導新的細胞株作為組織培養的新來源,就長遠角度來看利用枇杷細胞株生產有用物質方法,不僅具有保護植物資源、生產時間短、不受地區和季節限制、也沒有農藥污染的問題、達到經濟效益工業化生產等優點。此外,枇杷的三萜酸在最近藥理活性方面與各國相繼申請專利應用的商業價值,對於開發枇杷細胞株懸浮培養的方法於中草藥資源永續使用之研究發展上是值得開發利用之植物資源。

Study on Cell Suspension Cultures of Eriobotrya japonica Lindl

The aim of this study is development of the “resource of Taiwanese medicinal plant”. Using the loquat cell lines to establish the method of suspension culture and monitor the changes in the medium, as a base study to obtain a large scale of the secondary metabolites of bioreactor culture. In solid culture, it is not suitable for synchronization and monitor the medium composition ratio when culture. Furthermore, the limited space is unable to achieve a large scale of secondary metabolites. Thus, by means of the data on the previous study, we used the callus which cultured at the best culture condition to study the suspension culture of loquat. There are sufficient loquat seeds anytime in Taiwan and it is possible to induce callus as the new resource to establish new cell lines anytime. Moreover, there are many benefits to the production of useful secondary metabolites from loquat by suspension culture, such as protection of plant resource, rapid growth rate, no area and season limitation, agriculture pollution and economic benefit to industry. There are many bioactivities reports and patents of triterpenoids in loquat recently. Thus, to obtain large scale of secondary metabolites in loquat from suspension culture is valuable for traditional Chinese medicine research.
