促進植物細胞生物合成與指標成分的製備

在本實驗室已經建立的枇杷懸浮細胞培養的基礎上,探討枇杷細胞生合成三萜酸的各種前驅誘導物質或二次代謝物促激劑,一方面提高培養的細胞量、另一方面提高二次代謝產物三萜酸的含量,並且放大到35公升級的生物反應器進行培養。確立傳統藥材的生物技術利用,並提取純化培養細胞的主要有用成分,提供作為中藥指標成分之用。組織培養為高度技術與人力密集之作業。在傳統的三角錐形瓶培養中,每次操作所能生產的細胞量及二次代謝產物的效率很低,加上每次操作所投入的人力很高,其成本效益明顯不符合經濟效應。因此,科學家認為利用發酵槽/生物反應器是工業化中能有效的節省人力與降低成本技術、而且也是大量取得有用二次代謝產物三萜酸的重要關鍵技術。在本研究計畫中,將調控枇杷細胞的最適當生產二次代謝產物的條件,建立35公升的生物反應器的試生產培養技術,作為進入工業級產能的跳板,並進行培養細胞中有用物質蔷薇酸的純化與分離,提供作為中藥指標成分,以及藥物或中藥新藥開發的原料,確立生物技術在中藥材的永續資源利用。

Enhance of plant cell biosynthesis and separation of authentic sample

The aim of this study is development of the loquat cell lines to find the best method of suspension culture to synthesis second metabolites of triterpenoic acids, as a base study of precussor, elicitor to obtain a large scale of the secondary metabolites of bioreactor culture. Furthermore, use 35 liter bioreactor for a large scale of secondary metabolites syntehsis. Thus, to obtain large scale of secondary metabolites in loquat from suspension culture cells, And purify the bioavtive components for authentic sample of Chinese medicine.In the process of manufactory culture, it is not suitable for scale up suspension to use shake culture. Thus, by means of the data on the previous study, we used the callus which cultured at the best culture condition to study the suspension culture of loquat. And establish the 35 liter of culture technique to for manufactory process. Purify tormentic acid for authentic sample of Chinese medicine and there are valuable for traditional Chinese medicine research.
