常用中藥薏苡仁及其中成藥製劑對胚胎著床之安全性研究

薏苡仁是現代人常用的藥材與食材，中醫學上約略知道懷孕與月經週期的人不能食用薏苡仁，然而其科學上的機制仍不清楚，故利用科學的方法來研究其機制是必要的。本研究計畫的主要目標為運用生殖醫學的科學研究方法探討薏苡仁對胚胎著床之效應與可能的作用機制，以驗證中藥材薏苡仁在生殖醫學之毒理與藥效，來達到降低薏苡仁相關產品對生殖造成的副作用，提高薏苡仁在其他療效之方便性與安全性等。
本計畫主持人楊友仕教授的研究團隊專長於人類生殖醫學的研究。本研究計畫將以人類生殖醫學的研究方法配合薏苡仁食用及其中成藥製劑，探討薏苡仁對胚胎著床之效應並進一步運用薏苡仁的指標成分探討可能的分子作用機制。
主要研究策略與方法為：1.探討市面上常見的精白薏苡仁與糙薏苡仁及其中成藥製劑是否確實會造成活體流產：研究方法為運用胚胎植入技術以動物試驗探討餵食薏苡仁或方劑對胚胎著床及活產的效應。2. 探討薏苡仁造成活體流產的可能機制：研究方法為以動物試驗探討餵食薏苡仁或方劑對懷孕相關的內分泌分子及子宮內膜組織病理學的影響。檢測血清中E2、P4、oxytocin濃度，檢測免疫球(regulatory T cell)族群變化並以免疫組織染色檢測子宮內膜固著相關分子的變化。3. 探討薏苡仁指標成分對胚胎著床的影響：研究方法為運用體外體外胚胎著床細胞膜式，篩選出對BeWo球體附著有影響的薏苡仁指標成分 ，進一步探討特定薏苡仁指標成分對內膜細胞的生物效應。4.探討薏苡仁指標成分對活體內胚胎著床的影響：研究方法為為運用胚胎植入技術以動物試驗探討餵食特定薏苡仁指標成分對胚胎著床及活產的效應。並分別由血清學，免疫細胞與組織病理學的檢測釐清特定薏苡仁指標成分對內分泌，免疫調節及子宮內膜組織病理學的影響。以建立薏苡仁對胚胎著床效應之科學機制。
本研究計畫以系統生物學的概念由活體到分子機制探討中藥材薏苡仁與其指標成分在生殖醫學之效應，研究成果將能提供薏苡仁在藥用或食用上對於生殖醫學安全性的重要依據。此模式也將有助於建立中藥材在生殖醫學之安全性規範。

Elucidate the effect of Adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) seed and its traditional Chinese medicine on embryo implantation

Adlay (Coix lachryma-jobi L. var. ma-yuen Stapf) seed is a traditional Chinese medicine and as a nutritious food. Pregnant women are suggested not to eat adlay seed in the opinion of Chinese medicine. However, the scientific mechanisms of the adlay seed on embryo implantation are unclear. The major aim of this project is to elucidate the effect of adlay seed and its related Chinese herb reagents on embryo implantation and clarify the possible mechanisms of this effect. The results would help to establish the safety rules for the usage of adlay seed and its related Chinese herb reagents.
Professor Yang has focused on the field of human reproduction for several years; he has team up a study group that major in the technologies for in vitro and in vivo embryo implantation experiments. The study design and experimental methodologies of this project are: 1. to clarify the effect of commercialized adlay seed on embryo implantation by administration adlay seed and its related products with animal model. 2. To clarify the possible mechanisms of adlay seed and its related products on abortion by determining the their effects on the pregnancy related endocrine molecular, such as E2、P4、oxytocin in serum level, and check the population of regulatory T cell and the pathology of endometrial tissue. 3. To investigate the effect of indicator components and therapeutic components of adlay seed on embryo implantation with in vitro model. 4. To realize the effect of indicator components and therapeutic components of adlay seed on embryo implantation with in vivo model. The results would help to realize the safety of adlay seed and its related Chinese herb reagents on pregnant women and may help to establish the safety rules for the usage of adlay seed and its related Chinese herb reagents.
