應用蛋白質體學研究中藥茵陳蒿湯對保肝作用的分子調控機制(2-1)

隨著人類基因體解碼完成，伴隨而來的挑戰是探討蛋白質在生體中交互作用的生物功能角色。蛋白質體學是研究生物反應中受調控的蛋白質的有效方法，得到完整且具代表性的蛋白質圖譜是主要的關鍵，並可解決生物學與醫學的問題，提供了我們從整體系統的角度探討生物調節的複雜分子機制。 黃疸是人體組織內發生膽紅素聚積而造成的臨床表徵，新生兒黃疸尤其常見。目前針對新生兒黃疸除了紫外光照射外，以藥物為主的療法尚未普遍。臨床文獻報導中國大陸和日本使用中藥方劑茵陳蒿湯治療不同類型的肝疾病，發現其有利膽與保肝的特性。除此之外，實驗動物證據顯示茵陳蒿湯可以活化肝臟核受體(nuclear receptor)加速膽紅素的清除速率。藉由西方科學方法的印證來介紹中藥方劑對新生兒黃疸的可能療效，相對於傳統的光照療法不啻是一個令人新奇的經驗。 交互作用的蛋白質網路可以提供了解完整的細胞機制。研究功能性蛋白質體學的學者著重在蛋白質--蛋白質、基因--蛋白質的交互作用上。然而，有關中醫藥在功能性蛋白質體學的角色卻鮮少被提及。因此，本計劃將以蛋白質體學(Proteomics)來進行研究。找出茵陳蒿湯對實驗性膽汁淤積治療過程中肝臟、血清與肝細胞表現差異極大的蛋白質，利用”大量基因搜尋系統”(BGSS，http://servx8.sinica.edu.tw/bgss-cgi-bin/gene.pl)、蛋白質資料庫http://www.ncbi.nlm.nih.gov:80/entrez/query.fcgi/db=Protein/、http://www.expasy.ch/spro/、http://www.rcsb.org/pdb/，搜尋蛋白質資料，並將其作分類。以探討並嘗試建立茵陳蒿湯對實驗性膽汁淤積的療效機制圖譜。藉由本計劃的執行，我們可以較為完整的了解茵陳蒿湯對膽汁淤積的療效機制，同時提供未來對保肝中藥物開發的一個新方向。

Application of Proteomics on Hepatoprotection Effect of Chinese Herb Medicine Yin-Chen-Hao-Tang

As the genes of the Human Genome Project have finished, one of the challenges is to identify the biological function of the interactions between the whole human genes. Proteomics approach is a potentially powerful strategy for the discovery of regulated proteins in biological pathway. Therefore, to conquer a comprehensive protein map is a key factor for successful analysis. Using proteomics has provided an opportunity to investigate the complex molecular mechanism of biological processes. Jaundice, the accumulation of high levels of bilirubin in the circulation, is particularly common in human neonates. The treatment of choice for this condition in Western medicine is ultraviolet light therapy, but a drug therapy would also desirable. A Chinese herbal remedy for jaundice called Yin-Chen-Hao-Tang (YCHT) is now shown to activate a liver nuclear receptor that enhances the clearance of bilirubin. This herbal decoctions have been recognized as a magic bullet for jaundice and has long been used in China and Japan as a choleretic and hepatoprotective agent for various types of liver diseases.
