建立中藥化學指紋圖譜影像辨識之技術平臺

本計畫之目的為以影像辨識及模糊理論(fuzzy theory)為基礎,提供使用者一個快速、精準之中藥鑑別之技術平臺。首先利用溶媒對先行透過外觀與顯微辨識確認基原之中藥,進行萃取,並利用高效液相層析儀(HPLC)及核磁共振(NMR)技術,得到其參考標準圖譜,然後運用影像處理技術,抽取層析圖譜及NMR圖譜上的特徵資訊,並利用此資訊建構「中藥材層析指紋圖譜及NMR指紋圖譜影像資料庫」。就上述指紋圖譜影像採用從粗糙到細緻「Coarse to Fine matching」的比對模式進行辨識,在「Fine matching」階段則利用文獻所載及本研究室以鑑定之成分,建構多個歸屬函數(membership function)得到多個歸屬度(grade of membership),並以歸屬度做相似度之判別。而後,對未知或有疑問的藥材依本計畫所訂定之標準程式,就所取得之層析指紋圖譜及NMR指紋圖譜進行比對。此技術平臺可以藉影像辨識,確保使用藥材的規格能正確均一,進而使中藥製劑達到生產標準化的產品為目標,使中草藥製劑的療效與安全獲得保障。

To Construct an Image Recognition Platform to Identify the Fingerprint of Chinese Medicine

The aim of this project is to establish a rapid and accurate technique platform for identification of Chinese herbs based on image recognition and fuzzy theory. Firstly, Chinese herbs, whose origins being identified by morphology and microscopic examination will be extracted by solvents and the extracts will be analyzed by high performance liquid chromatography (HPLC) and nuclear magnetic resonance spectroscopy (NMR) to obtain the HPLC and NMR fingerprints, respectively. The characteristic information of HPLC and NMR fingerprints will be extracted through the technique of image treatment, and the image libraries of HPLC and NMR fingerprints of herbs will be thus established. The fingerprint imagines obtained will be recognized by the comparison model using coarse to fine matching. In the fine matching stage, multiple membership functions will be constructed for constituents of Chinese herbs, which have been reported in the literature or identified previously in our laboratory, and multiple grades of membership will be obtained. Then, the degree of similarity will be determined according to the multiple grades of membership in the fingerprints. The present technique platform of imagine recognition will be used to clarify unknown or suspected Chinese herbs. It is anticipated that misuse of Chinese herbs will thus be avoided and the quality assurance of herbal preparations will also be provided.
