利用免疫細胞吞噬活性為快速篩檢平台評估具抗發炎與增強免疫力之中草藥

免疫系統常誘導發炎反應來對應外來病原,並誘使防禦細胞前來,其參與的免疫細胞包括單核球與其特化之巨噬細胞、顆粒球與自然殺手細胞等,其防禦過程中在免疫細胞與其分泌性循環性因子的交互作用下,由防禦細胞的吞噬與消化作用,來清除外來病原與受損細胞。我們利用吞噬細胞活性為篩檢平台來評估市面上聲稱具有增強免疫及抗發炎之中草藥,利用人類與小鼠巨噬細胞株進行體外培養之篩選模式以中草藥之萃取液與培養基共培養之模式,評估吞噬細胞活力與細胞激素分泌之調節,並探討其可能之分子機轉。吞噬活力以流式細胞儀分析巨噬細胞吞噬螢光標示之大腸桿菌(E. coli, K-12 strain)或Latex bead之能力,並分析巨噬細胞活化過程中對於一氧化氮之產生與鈣離子於細胞質的增加作為評估工具,並評估吞噬細胞表面分子Toll-like receptor (TLR)的變化。本計畫中使用流式細胞儀作為快速分析之工具,首先以巨噬細胞株培養模式快速篩選對於吞噬活力與細胞激素、鈣離子、NO與TLR表現產生調節的適當單味藥,進一步使用篩選後之單味藥或適當配伍來分析動物模式中經病原活化之吞噬細胞活力與細胞激素之分泌調節,或許可最為臨床前試驗或臨床應用之參考。

Immune system used the inflammatory response to against the pathogen. The cells involved in this inflammatory response include macrophage, neutrophil and nature killer cell. The phagocytes play the central role in the clearances the pathogen and damaged cells. In this study, we will use the phagocytosis to monitor the immunomodulated effect in herbal medicine so called “improved immune response and anti-inflammation herbs”. We will use human or murine macrophage to phagocytosis the fluorochrome-labeled E. coli (K12 strain) or latex bead to monitor the ability of phagocytosis. In this response, nitric oxide, calcium influx and pro-inflammatory cytokines will be monitored to inflammation. Further effected herbs will be used in the LPS induced acute lung injure in murine model to monitor the immune response including pulmonary macrophage phagocytosis and inflammation.
