厚朴對熱中風動物模式療效之評估

我們過去的研究報告指出熱中風發生時,動物的動脈壓下降、單胺濃度昇高、顱內壓昇高、腦局部血流減少、神經細胞損傷,以致於動物的存活期縮短至十五分鐘左右。如果事先將動物腦中的單胺(尤其是妥巴胺或5- 色胺)濃度降低,則可以延長熱中風動物的存活期,並減輕熱中風動物的低動脈壓、高顱內壓、腦缺血及細胞損傷等症狀。最近我們也發現中藥成份厚朴酚具有很強的降低動物腦內單胺物質濃度的作用,因此我們擬在本實驗中探討厚朴酚對於熱中風動物模式之治療效果。首先,我們建立熱中風動物模式;將全身麻醉的大鼠暴露置於高溫環境(42oC)以確定熱中風發生時出現低血壓、高顱內壓、腦缺血、細胞損傷及縮短存活率等現象。然後觀察當給予厚朴酚治療後,上述熱中風症狀群是否被改善。從本實驗預期的結果,可以得知厚朴酚對於熱中風動物模式之療效有多大,且經由何種機制來達其療效。本實驗預期之成果,不僅可在熱中風生成的學理上有所建立,並且可提供熱中風臨床治療上的參考依據。

Therapeutic evaluation of magnolol on heatstroke animal model

Our previous results have shown that the animals with heatstroke displayed arterial hypotension, intracranial hypertension, cerebral ischemia, neuronal damage and shorted survival. Depleting monoamines (in particular the dopamine and serotonin) in the brain with neurotoxin significantly extended the survival of the animals with heatstroke and attenuated the heatstroke-induced arterial hypotension, intracranial hypertension, cerebral ischemia, and neuronal damage. Recently, our results also demonstraated that the monoamine levels of rat brain were greatly decreased after systemic administration of magnolol. Therefore, the present study will assess the effects of systemic administration of magnolol on the survival of the heastroke animals and the heatstroke-induced arterial hypotension, intracranial hypertension, cerebral ischemia and neuronal damage.
