減肥中草藥之藥效與肝腎毒性的評估(2-2)

本計畫利用兩年的時間,針對市面上常用於減肥的中草藥藥材炮製品,評估是否具有預防高熱量飼料引至實驗動物罹患肥胖症之效果,同時針對長期服用這些待測中藥及其炮製品後是否可能引至肝毒性、腎毒性進行評估。第一年度計畫已完成具有改善高血脂作用的何首烏與柴胡以及藥材炮製品,製首烏與醋柴胡的調控體脂不易形成功效與可能引至肝腎副作用評估的結果。第二年度計畫將針對具有利尿作用的澤瀉和決明子,與其藥材炮製品,分別為鹽澤瀉與炒決明子進行探討。本年度計畫將藉由每星期檢測高熱量飼料引至肥胖大鼠體重的變化,配合白色脂肪細胞的體積與數目的觀察結果與肝臟氧化體增殖活化受體PPARα與脂質氧化酵素ACO蛋白的表現量評估待測藥材與其炮製品調控體脂不易形成功效與可能藉由的作用機轉。另外,藉由血液中肝功能生化指數glutamic pyruvic transaminase (GPT)與glutamic oxaloacetic transminase (GOT)的變化以及血漿肌酐酸(serum creatinine)、血尿素氮(blood urea nitrogen)濃度的檢測,配合肝臟與腎臟的病理切片評估結果,探討待測中藥材及其炮製品對實驗動物肝臟、腎臟傷害的程度。本項計畫的完成將有助於證實這些待測藥材與其炮製品是否具有預防或是改善高熱量飼料引至實驗動物罹患肥胖症之效果,並找出可能產生療效的劑量。此外可瞭解長期服用這些待測中藥及其炮製品後是否可能引至肝毒性與腎毒性。所得結果將可提供產官學各界有關中藥服用安全評估、政策的建立及研究方向的參考。

Evaluation on the anti-obesity efficacy as well as kidney and liver toxicity induced by Chinese medicinal plants (2-2)

The research is the two-year plane. The aim of the research is going to evaluate the effects of herbal medicines and the processed products that commonly used in weight reduction, on prevention of high energy diet-induced obesity in rats. In addition, the possible hepatotoxicity or nephrotoxicity induced by long-term administration of these anti-obesity herbal medicines and its processed products will be evaluated. In the first year, anti-obesity efficacy and both hepatotoxicity and nephrotoxicity associated with herbal with hypolipidemic activity, such as Polygoni Multiflori Radix and its black bean sauce-processed product, as well as Bupleuri Radix and the vinegar-processed product, have been evaluated. In the second year, the anti-obesity efficacy and the liver and kidney toxicity caused by herbal diuretic including Alismatis Rhizoma and salt-processed product, or Semen Cassiae and firing-processed products, will be investigated. During the experimental period, the changes in body weight will be evaluated weekly. The volumes and the numbers of white adipocytes as well as the hepatic protein levels of peroxisome proliferator-activated receptor (PPAR) α and acetyl-CoA oxidase (ACO) will also be investaged to clarify the efficacy and the action mechanisms of the tested herbal medicines and its processed products on the controlling of body fat formation. The plasma levels of glutamic pyruvic transaminase and glutamic oxaloacetic transminase will be determined to evaluate whether the hepatotoxicity will be induced by the tested herbal medicines and the proceed products. Kidney damage induced by the tested herbal medicines and the processed products will be determined by the levels of blood urea nitrogen and plasma creatinine concentration. The biopsy will be further performed to evaluate the pathology of hepatotoxicity and nephrotoxicity. The results will provide valuable information about safety of tranditional Chinese medicine for both academic and industrial parties and maybe helpful for the establishment of related policy.
