加味六君子湯在腫瘤小鼠模式中的抑癌療效評估及機轉探討

癌症一直高居台灣十大死因之首。癌症治療方式主要有手術、化學治療、放射線治療。而癌症免疫療法近年來亦逐漸受到重視。雖然這些治療方法可以提高病患存活率,但副作用及復發率都是迫切需要解決的問題,所以發展更好的抗癌策略是十分必要的。近年來中藥及其成份應用於癌症治療或輔助現有之癌症治療方法已逐漸被重視。其中扶正固本藥方被視為可降低化放療的毒副作用,亦可強化骨髓造血機能進而增進病人免疫力,降低腫瘤生長及復發。我們的合作者市立聯合醫院許中華醫師根據其臨牀經驗以扶正觀點,發展出一扶正中藥複方(加味六君子湯),由黨參、白朮、茯苓、甘草、黃耆、藿香、女貞子等7種中藥組成,其中數種中藥已被報導與抗癌及增強免疫有關。本實驗室的初步結果發現此方在原生性小鼠膀胱癌模式中可降低腫瘤生長且無明顯毒性。本計畫將進一步探討加味六君子湯的抗癌作用。我們將研究加味六君子湯抑制膀胱癌小鼠腫瘤的作用機轉,也將評估此方在肺癌小鼠動物模式中是否對腫瘤也有抑制作用。另外,也將測試加味六君子湯和化療並用後是否可增強抗癌效果並降低化療所誘發之毒性。

Cancer is the leading cause of death in Taiwan. At present, conventional therapies for cancer include surgery, chemotherapy and radiotherapy. Recently, cancer immunotherapy also draws a lot of attention. Although these therapeutic strategies can improve survival rate of cancer patients, therapy‐induced adverse effects and cancer relapse limit therapeutic outcome. Effective cancer treatment strategy with less side effects is still in great demand. Recently, using herbal medicine or its derived components as cancer therapeutic agents or as adjuvant for conventional therapy are getting much acceptance worldwide. Particularly, herbal medicine that reinforce the healthy qi are applied to alleviate the side effects of chemo/radiotherapy, enhance immunity and delay cancer growth and reoccurrence. Based on the concept of “reinforce the healthy qi”, our collaborator Dr. Chung‐Hua Hsu has developed a compound herbal formula (modified Liow‐jiun‐Tzyy‐Tang), which contains seven herbs including Radix codonopsis, Atractylodes Rhizoma, Poria cocos, Radix Glycyrrhizae, Radix astragali, Herba pogostemonis and Fructus Ligustri Lucidi. Several herbs in this compound herbal formula are previously reported to exhibit anti‐cancer and immunomodulatory effect. Furthermore, our preliminary data showed that modified Liow‐jiun‐Tzyy‐Tang delayed tumor growth without causing any toxicity. This project will further investigate the anticancer effects of modified Liow‐jiun‐Tzyy‐Tang. We will investigate the action mechanism underlying tumor regression induced by modified Liow‐jiun‐Tzyy‐Tang. We will also examine whether modified Liow‐jiun‐Tzyy‐Tang also exhibit antitumor activity in lung cancer mouse model. Finally, we will investigate whether combined treatments of odified Liow‐jiun‐Tzyy‐Tang and chemotherapy have synergistic effects in tumor egression and could reduce the chemotherapy‐induced toxicity.
